The historical Takuan Soho was a genuine figure: a Rinzai Zen monk, calligrapher and advisor who lived through the same era as Musashi and is associated with the development of Zen-influenced swordsmanship thought. A Zen monk positioned against a wandering swordsman, both carrying the same demonic hand, is a promising setup.
Hatsugumo Okuno
Hatsugumo Okuno is described as a warrior among the notable characters in the game. Beyond that, published detail is thin, and we are not going to invent a backstory to fill the space.

The people of Kyoto
One group deserves a mention even though they are not named characters. Preview coverage describes a town where effectively every resident has made a bargain with the Genma, trading parts of their bodies to have their problems solved.
The example given in coverage is grimly memorable: a woman who must keep playing an instrument, and who is punished when she plays it wrong, is offered a fix by a Genma — it will simply take her fingers. She agrees. The game treats this as a horror-tinged deal with the devil rather than a joke.
These residents are how the story establishes what the Genma do to a city, and they are why the objective in that section is to find the Genma collecting the body parts rather than simply clearing enemies.
